From simple links to polygenic complexity
Early research in the 1990s suggested a correlation between the MAOA gene variant and violent tendencies. By 2004, media had popularized the term 'warrior gene,' framing it as a direct biological driver of aggression. However, contemporary science paints a far more nuanced picture.
Aysu Okbay, assistant professor of psychiatry and complex trait genetics at Amsterdam UMC, notes that the once-dominant belief in a few genes exerting major behavioral effects has been 'completely debunked.' Even traits like height, long considered highly heritable, resist simple genetic mapping.
Twin studies and the limits of heritability
The 'nature or nurture' debate gained traction in the 19th century through Francis Galton's twin studies, though modern methods emerged in the 1920s. Today, personality is often measured through the 'Big Five' traits: openness, conscientiousness, extraversion, agreeableness, and neuroticism.
A 2015 meta-analysis of 2,500 twin studies found that genetic differences account for roughly 47% of personality variations, with environmental factors explaining the rest. Yet identical twins-who share 100% of their DNA-rarely exhibit identical personalities, underscoring the complexity of inheritance.

Genome-wide studies and the 'missing heritability'
Advances in genome-wide association studies (GWAS) have revealed that personality traits are 'polygenic,' influenced by thousands of genetic variants, each contributing minimal effects. While twin studies suggest 40-50% heritability for personality, GWAS estimates range from 9% to 18%.
Okbay attributes this discrepancy to small sample sizes and incomplete understanding of gene interactions. 'It's probably somewhere between the two,' she says, emphasizing the need for larger, more diverse studies. Daniel Levey of Yale University adds that current research disproportionately focuses on European ancestry, potentially overlooking cultural variations.
Environmental influences: Beyond trauma and upbringing
While major life events-like winning the lottery or experiencing trauma-are often assumed to reshape personality, studies show their impact is negligible. Childhood adversity, however, can predict higher neuroticism and cognitive challenges later in life. Brent Roberts of the University of Illinois notes that adult trauma leaves 'no huge trace,' challenging popular narratives of personal growth through suffering.
Prenatal conditions may also play a role. A 2022 study linked maternal stress during pregnancy to increased fear and distress in infants, though the mechanisms-possibly epigenetic-remain unclear. Personality differences are now understood as 'poly-environmental,' with myriad small experiences cumulatively shaping behavior.
Emerging insights and unresolved questions
Recent GWAS involving hundreds of thousands of participants have identified hundreds of DNA variants tied to the Big Five traits. Levey's research highlights CRHR1, a stress-response gene linked to neuroticism and psychiatric disorders. Another forthcoming study suggests the prefrontal cortex-a brain region tied to decision-making-hosts genetic associations for most Big Five traits, except agreeableness.
Yet even well-studied areas, like the 'warrior gene,' remain contentious. While certain genetic and environmental risk factors (e.g., childhood abuse) may increase aggression in some males, findings are inconsistent. Jana Instinske of Bielefeld University stresses that genetic predispositions are not deterministic: 'Behavior depends on environment and context.'
The mutable human condition
Decades of research reveal that personality arises from a dynamic interplay of genetics, environment, and chance. As Okbay notes, 'We're only beginning to detect these tiny effects.' The path forward lies in larger, more inclusive studies-and a recognition that human complexity defies reduction to a handful of genes or life events.
